ABSTRACT

The objective was to evaluate the antioxidant capacity, glucose and insulin concentration and reproductive performance of ewes supplemented with orange residue prior to insemination. Fifty-five multiparous ewes were divided into two corrals, and 15 unbred ewes were kept individually to measure feed consumption. Two integral diets were administered; T0: control treatment and T1: with 20% of dry matter of orange residue. Ten days after the start of supplementation, the ewes were synchronized. Supplementation was finalized prior to artificial insemination, then, a blood sample was taken to measure the antioxidant capacity and glucose and insulin concentration. An analysis of variance was made to evaluate the effect of treatment on the antioxidant capacity, glucose and insulin; and to analyze the response to estrus, percentage of gestation and prolificity a ji squared test was performed. Of 9 antioxidant compounds found in the orange residue, hesperidin (7.44%), chlorogenic acid (0.50%) and protocatechuic acid had the highest concentration. Feed intake, estrus response, percentage of gestation, antioxidant capacity, and glucose and insulin concentration were not affected by the treatment. It is concluded that inclusion of 20% of orange residue in the diet prior to insemination in ewes is possible.(AU)

ABSTRACT

The objective of this study was to compare the reproductive efficiency of dairy buffaloes undergoing fixed-time artificial insemination (FTAI) protocols based on progesterone/estrogen (P4/E2) and eCG during unfavorable breeding season using cooled (CS) and frozen semen (FS). A total of 446 buffaloes (> 40 days postpartum) were randomly distributed into four blocks (years): B1-2014 (n = 143), B2-2015 (n = 34), B3-2016 (n = 90), and B4-2017 (n = 179). Each block was subdivided into two (AI with CS and FS using the same ejaculate of each bull). Thus, the block subdivision was as follows: B1 (CS = 71 and FS = 72); B2 (CS = 18 and FS = 16); B3 (CS = 47 and FS = 43); and B4 (CS = 90 and FS = 89). The ejaculates of eight Murrah bulls collected using an artificial vagina were divided into two aliquots: one aliquot was diluted in Botu-Bov® commercial extender and cooled (BB-CS), and the other was diluted in the same extender and frozen (BB-FS). BB-CS aliquots were cooled at 5 °C/24 h using a refrigerator. BB-FS group aliquots were also cooled, and after equilibrating at 5 °C for 4 h, were placed in a 21-L Styrofoam box, 5 cm above the surface of liquid nitrogen. In the afternoon (A) on D0 (2:00 p.m.) the animals received EB 2.0 mg IM (Estrogin®) and an ear implant (CRESTAR® 3.0 mg P4). At D9 (A), the implant was removed, and the animals received eCG 400 IU IM (Folligon® 5000) + Cloprostenol PGF2α 0.530 mg IM (Sincrocio®). At D10 (A), the animals received EB 1.0 mg IM (Estrogin®), and at D12 (8:00 a.m.), AI was performed. At D42, pregnancy was diagnosed via ultrasonography. Total CRs were 48.2% CS and 34.6% FS for years 2014 to 2017, with a significant difference of 13.7% (P<0.05). In conclusion, cooled semen resulted in higher CR than frozen semen in dairy buffaloes under the P4/E2 and eCG FTAI during the unfavorable reproductive season.(AU)

Assim, a subdivisão do bloco foi a seguinte: B1 (SR = 71 e SC = 72); B2 (SR = 18 e SC = 16); B3 (SR = 47 e SC = 43); e B4 (SR = 90 e SC = 89).

ABSTRACT

The objective of this study was to investigate the influence of plasma pathological changes before timed artificial insemination (TAI) on pregnancy of cows. The contents of estrogen (E2), progesterone (P4), glucose (Glu), selenium (Se), brain-derived neurotrophic factor (BDNF), and histamine (HIS) in plasma of 48 Holstein cows were measured before TAI. According to the estrus detection, the cows were divided into estrus (E) and anestrus (A) groups. After pregnancy testing at 28 d after TAI, two groups of E and A were divided into positive pregnancy of E group (EP+), negative pregnancy of E group (EP-), positive pregnancy of A group (AP+), and negative pregnancy of A group (AP-). The contents of E2, P4, Glu, Se, BDNF and hIS significantly differed among the four groups (P<0.01). The ROC analysis was used to determine the risk of negative pregnancy test (-) after TAI was increased when plasma E2 was less than 46.45 pmol/L in cows before TAI. The changes in E2, P4,hIS, Glu, and BDNF in the blood of natural estrus and natural anestrus cows affected the pregnancy after TAI. the level of E2 in plasma may be used to assess the risk of negative pregnancy after TAI.(AU)

ABSTRACT

The present investigation was carried out with the total of 18 Sahiwal cattle of 1st to 4th parity were selected for the study and distributed into three different groups based on their pre calving BCS (before 15 days of expected date of calving) namely G1 (2.50 - 3.00), G2 (3.25 - 3.75) and G3 (4.00 and above) with six animals in each group. The average fortnight milk yield increased up to 60th day and thereafter declined up to 90th day in G1, G2 and G3 group of animals, which was significant (p<0.05) up to 75th day. G3 group animals had highest average fortnightly cumulative Milk yield (Kg) in entire study period. Highest 305 days predicted lactation milk yield (2462.66 Kg /lactation) and average peak yield (16.50 Kg/ animal) was obtained in G3 group animals and the lowest predicted lactation milk yield (1870.68 Kg/lactation). The 305 days predicted lactation milk yield in G2 group animals was 2356.75 Kg /lactation and average peak yield was 14.90 Kg/ animal. The G3 group animals also attained their peak yield in 46 days compared to G2 and G1 group animals with 51 and 54 days, respectively. The persistency of milk production was highest in G2 group animals with 65.63 % followed by G3 and G1 group animals. The studied revealed that BCS had a significant (P<0.05) effect on post-partum estrus and service period in experimental animals

ABSTRACT

The present study was aimed on the genetic improvement of non-descriptive goats through estrus synchronization and by crossbreeding with Beetal goats they were reported to be a good milker, having higher body weight and dressing percentage. AVIKESIL-S, indigenously developed progesterone intra-vaginal sponges were utilized for estrus synchronization in does maintained at livestock farm complex, Veterinary College, Hassan. Response to estrus synchronization was noticed in all the does (100%) under study, irrespective of the genetic background. Natural service was practised with three mating designs ie. G1-Beetal × Beetal, G2-Beetal × non-descriptive, G-3 non-descriptive × non-descriptive. Pregnancy test was performed with ultrasonography at 45 days post-mating. The conception rates in does of G1, G2 and G3 groups were 87.5, 87.5, and 75.0% respectively. Kidding rate were 85.7, 85.7 and 100.0% in G1, G2 and G3 groups, respectively. Litter sizes were 1.5, 1 and 1 in G1, G2 and G3 groups, respectively. Average birth weight in G1, G2 and G3 groups were 3.82±0.20, 2.82±0.12 and 2.00±0.09 kg, respectively and average daily gain in G1, G2 and G3 groups were 42.86±1.69, 33.81±1.28, and 33.57±0.87 g, respectively. Better growth performances were noticed up to 3 months of study period in G1 and G2 compared to G3 groups. It could be concluded that combined utilization of ES technology and crossbreeding in non-descriptive goats would provide superior kids for bulk marketing and ease management by avoiding dispersed kidding. Also more number of animals would be available for selection thus increasing the selection intensity their by improving the genetic improvement

ABSTRACT

The present study evaluated the effect of circulatory trace mineral profiles on fertility response during estrus induction by progesterone based therapy in acyclic Sahiwal cattle. Sahiwal cows and post-pubertal heifers (n=25 each) with true anestrus condition (acyclicity confirmed ultrasonographically) were subjected to CIDR-based protocol (intravaginal CIDR application for 7 days, 500 IU eCG at CIDR removal, followed by estrus observation and AI). In heifers, additionally an injection of 5.0 mg estradiol valerate at CIDR insertion was given. Blood samples on days 0 (before CIDR application), 3, 5, 7 and 9 were collected. Almost similar estrus induction (68% vs. 64%) and subsequent conception rates (41.2% vs. 50%) during the first induced estrus were recorded in cows and heifers, respectively. During the course of treatment, significant (P<0.05) variation in plasma copper and zinc concentrations was recorded both in cows and heifers. Contrarily, manganese concentrations varied significantly (P<0.05) in cows only. However, iron profiles remained uniform during the course of treatment. Significantly (P<0.05) higher pre-treatment plasma concentrations of copper, zinc and manganese was recorded in those cows and heifers in which estrus was induced, compared to those in which estrus induction failed to occur. No significant difference (P>0.05) was recorded for iron concentration vis-à-vis estrus induction response. Hence, it is indicated that circulatory copper, zinc and manganese profiles influence the reproductives energy in acyclic animals and their suboptimal profiles may reduce the efficacy of estrus induction protocols in Sahiwal cattle.

ABSTRACT

Abstract Background: Coffee pulp has a high content of antioxidants capable of modifying the oxidative status in small ruminants. However, high amounts for a prolonged time can reduce fertility. Objective: To determine the effect of two inclusion levels of coffee pulp during estrous synchronization on reproductive variables and oxidative status of primiparous ewes. Methods: Sixty Suffolk x Dorset primiparous ewes were distributed into three treatments in a completely randomized design; T0: (n=20) 1.5 kg balanced diet, T1: (n=20) 1.5 kg balanced diet and 5% coffee pulp, T2: (n=20) 1.5 kg balanced diet and 10% coffee pulp. Supplementation was given for 16 days before estrus synchronization and until the beginning of the breeding season. A progestogen (CIDR®) was inserted for 11 days and a dose of PGF2α was applied two days prior to its withdrawal. Estrus detection started 12 hours after CIDR withdrawal. Blood samples were obtained during the supplementation period to measure oxidative status, antioxidant capacity, glucose and insulin, and up to 9 days after breeding to determine progesterone concentration. Pregnancy diagnosis was performed at 30 and 60 days post-breeding. An analysis of repeated measures of mixed effects and frequency analysis were carried out. Results: Inclusion of coffee pulp for a short period prior to breeding did not affect reproductive parameters, nor progesterone, glucose or insulin concentrations (p>0.05); however, antioxidant capacity increased, while lipid oxidation showed an opposite trend (p<0.05). Conclusion: Inclusion of up to 10% coffee pulp in the diet of ewe lambs for 16 days prior to breeding improves oxidative status without causing adverse effects on pregnancy, estrus or prolificacy.

ABSTRACT

This study aimed to obtain the electrophoretic profile of serum proteins in cyclic mares and to verify the differences between the follicular and luteal phases of the estrous cycle in this species. Eighteen mares were used, totaling 36 serum samples, two of each mare. Samples were collected both in estrus and in diestrus. Total serum proteins were obtained by the Biureto method, by using commercial kits (LABTEST), while the different protein subfractions by polyacrylamide gel electrophoresis (SDS-PAGE). The electroforetogram of serum proteins evidenced the presence of 17 to 25 protein fractions, whose molecular weights ranged from 22 to 254 kDa. Two proteins that were not yet officially named were identified, of molecular weights (MW) of 23 kDa and 144 kDa. The mean values (± SEM) obtained for each variable in estrus and diestrus were, respectively: total proteins (g/dL) 7.11 ± 0.07 and 7.36 ± 0.07; albumin (mg/dL) 4790.83 ± 69.10 and 5027.19 ± 69.10; α1 acid glycoprotein (mg/dL) 4.90 ± 0.31 and 4.93 ± 0.31; ceruloplasmine (mg/dL) 15.28 ± 1.31 and 10.65 ± 1.31; haptoglobine (mg/dL) 22.70 ± 1.16 and 27.06 ± 1.16; transferrin (mg/dL) 329.00 ± 9.78 and 350.16 ± 9.78; IgA (mg/dL) 119.91 ± 6.30 and 107.03 ± 6.30; IgG (mg/dL) 1525.07 ± 40.18 and 1517.25 ± 40.18; MW 23 (mg/dL) 204.44 ± 8.61 and 219.79 ± 8.61; MW 144 (mg/dL) 22.13 ± 0.55 and 21.49 ± 0.55. No significant difference was verified in total proteins and its fractions in estrus and diestrus. The hormonal changes during the specific stages of the estrous cycle of the mare do not interfere with the serum proteinogram.

ABSTRACT

Abstract The use of progestogens can contaminate the environment and animal tissue. Therefore, the use of prostaglandins F2α (PGF2α) and gonadotropin-releasing hormone (GnRH) may represent a viable alternative. The aim of the present work was to evaluate the effect of applying a dose of GnRH within the range of application of two doses of PGF2α, and compare it with protocols based on progestogens and PGF2α, commonly used in the synchronization of estrus, with respect to the pre-ovulatory peak of luteinizing hormone (LH), post-estrus progesterone concentrations, pregnancy rate, calving and prolificacy. After a pre-synchronization with a double application of PGF2α, 30 multiparous adult ewes (live weight = 45 ± 3.1 kg, body condition = 3.2 ± 0.5 units and postpartum time = 60 d to 90 d, were randomly assigned to one of three treatments (n = 10). The ewes in Group 1: P (Control) received a single dose of 125 μg of cloprostenol via IM (intramuscular); the ewes in Group 2: FP received intravaginal sponges containing 20 mg of fluorogestone acetate for 5 d; and upon removal of the sponge, they were injected with 125 μg of cloprostenol via IM; the ewes in Group 3: GP received 50 μg of GnRH IM, and 7 d later they were injected with 125 μg of cloprostenol. The onset of estrus was detected faster (P ≤ 0.05) in P and GP groups than in the FP group. The GP group (P ≤ 0.05) had higher concentrations of progesterone after 5 d post-estrus, compared to the P and FP groups. The pregnancy rate was not different (P > 0.05) between groups. All the synchronization protocols evaluated were similar in the effectiveness to improve the reproductive behavior in hair sheep. Therefore, the use of protocols based on the application of PGF2α is recommended, due to their its ease of application.

ABSTRACT

ABSTRACT

Abstract This research aimed to develop a surgical technique to prepare teaser rams. This was proposed because the techniques practiced in the Colombian savanna of the Córdoba region have not shown satisfactory results, since in the postoperative period sheep present difficulty to heal and long convalescence, which has a considerable economic cost. For this reason, many owners opt for sacrificing the animal, which also increases the losses of producers. For this work, a group of ten clinically healthy Creole sheep was used, with an average age of 1 year and an average weight of 21 kg. For the surgery, sheep were tranquilized with intravenous acepromazine maleate at a dose of 0.02 mg/kg, and anesthesia was achieved with the epidural injection of 2% roxicaine without epinephrine in the dorsal intercoccygeal region (Co2-Co3) at a dose of 1 mL/5 kg of live weight. After successfully blocking the area, the surgical technique was performed, which lasted an average of 3 min. The operated animals were evaluated during 6 months; good healing and ability to urinate was observed. As for libido, all had a positive behavior in front of females in heat. 100% of the animals were unable to penetrate the females; only 10% of them had partial erection of the penis (glans). The proposed technique was effective, non-invasive, quick, easy to perform, and economical. It does not require specialized instrumentation and does not have postoperative complications, which allows using sheep as teaser animals at 15 days post-operation.

ABSTRACT

We compared the efficiency of protocols for estrus induction in agoutis. Five females received double intraperitoneal administration of cloprostenol (5µg) on a 2-days interval; other five females were treated with intravulvar administration of 30µg gonadotrophin release hormone analogue (GnRH associated to intraperitoneal administration of 5µg cloprostenol after seven days and a new administration of GnRH analogue after two days. Every 3 days, the agoutis' reproductive cycle was monitored by blood collection for hormonal analysis, ovarian ultrasound and vaginal cytology. Two females, originally in luteal phase, that received isolated prostaglandin presented estrous signs at 3 and 6 days after the second drug administration. From the females that received the association, two that were originally in luteal phase presented estrus at 4 days after treatment, and one other presented estrus only after 10 days. There was no significant statistical difference regarding the efficiency of treatments for estrus induction (P>0.05). We conclude that, according to the protocols tested in the study, the use of isolated prostaglandin or its association to GnRH analogue for estrus induction in D. leporine shows an efficiency limited to the females that were in luteal phase in the beginning of the treatment.(AU)

ABSTRACT

Abstract Ovarian cysts are common anomalies in dairy cattle. They are characterized by presenting with anestrus, irregular estrus, increased reproductive intervals, and infertility. Their origin is still unknown, thus it is important to study their pathogenesis and possible relation to certain factors predisposing to their onset. The objective of this study was to determine by ultrasound the prevalence and types of ovarian cysts. 93 cows belonging to 23 farms in the Hoya de Loja were studied. Cystic formations were identified and classified according to ultrasound profile during 24 days, with 6 scans every 4 days. The age of the animals varied from 2 to 12 years, with body condition > 2. For data analysis, PROC REG was used in the SPSS Statistics 21 statistical program. An increase of 256.4 open days was evidenced in cows with ovarian cysts, versus 114.8 open days in healthy control animals. The prevalence of ovarian cysts was 32%, distributed in follicular cysts (21%) and luteal cysts (11%). In addition, there was a correlation between the presence of cysts and body condition < 3 (23%), presence of metritis (9%), and the use of hormonal products (10%). Ovarian cysts were clinically manifested through symptoms of anestrus in cows with luteal cysts (90%) and the presence of irregular estrus in cows with follicular cysts (40%). In conclusion, ovarian cysts are a direct cause of increased periods of delivery-estrus and delivery-conception, as well as increased intervals between deliveries in postpartum dairy cows.

ABSTRACT

Background: The effect of aqueous stem bark extract of Anogeissus leiocarpus (AEAL) on the reproductive cycle of adult female Wistar rat and the serum levels of ovarian hormones (estradiol and progestin) was studied. Methods: Twenty-four (24) adult female Wistar rats were used. Rats were divided into groups (I - IV; n=6). Group I was control and received distilled water; groups II, III and IV received AEAL (200mg/kg, 400mg/kg, and 600mg/kg, oral, respectively) for the period of six (6) weeks. The estrous cycle changes were determined by daily observation of vaginal smear, while serum levels of estradiol and progestin was compared after the extract administration-using enzyme linked immunosorbent assay kits. Results: Result showed significant (p< 0.05) increase in the levels of serum progesterone and estradiol in groups II and III treatment; 19% higher when compared to the control. Treatment with AEAL at doses 200mg/kg and 400mg/kg prolonged di-estrus and estrus phases of the rats’ estrous cycle. The increase in the duration of the two phases (diestrus and estrus) was dose dependent, significant (p<0.05) when compared to the control. Conclusion: AEAL has effect on the reproductive cycle and serum levels of ovarian hormones of Wistar rats, hence of potential use in fertility related studies.

ABSTRACT

OBJECTIVES: To evaluate the effect of Carbopol gel formulations containing pilocarpine on the morphology and morphometry of the vaginal epithelium of castrated rats. METHODS: Thirty-one female Wistar-Hannover rats were randomly divided into four groups: the control Groups I (n=7, rats in persistent estrus; positive controls) and II (n=7, castrated rats, negative controls) and the experimental Groups, III (n=8) and IV (n=9). Persistent estrus (Group I) was achieved with a subcutaneous injection of testosterone propionate on the second postnatal day. At 90 days postnatal, rats in Groups II, III and IV were castrated and treated vaginally for 14 days with Carbopol gel (vehicle alone) or Carbopol gel containing 5% and 15% pilocarpine, respectively. Next, all of the animals were euthanized and their vaginas were removed for histological evaluation. A non-parametric test with a weighted linear regression model was used for data analysis (p<0.05). RESULTS: The morphological evaluation showed maturation of the vaginal epithelium with keratinization in Group I, whereas signs of vaginal atrophy were present in the rats of the other groups. Morphometric examinations showed mean thickness values of the vaginal epithelium of 195.10±12.23 μm, 30.90±1.14 μm, 28.16±2.98 μm and 29.84±2.30 μm in Groups I, II, III and IV, respectively, with statistically significant differences between Group I and the other three groups (p<0.0001) and no differences between Groups II, III and IV (p=0.0809). CONCLUSION: Topical gel formulations containing pilocarpine had no effect on atrophy of the vaginal epithelium in the castrated female rats.

ABSTRACT

Objective To provide ideal endometriosis animal models for research new treatment methods. Methods 64 Sexual mature without pregnant SD rats who has regular oestrous cycle were underwent operation that autologous endometrium were transplanted to peritoneum and subcutaneous with lancet gently cut tissue of where to transplant in the rut, and compared the model of peritoneum and subcutaneous after 4 weeks. Results The general success rate of autologous endometrium transplantation in rat estrus was 93.3%,and the peritoneum transplantation success rate was 51.7%, subcutaneous was 88.3%, the difference of the two place transplantation has statistical significance. Compare the two parts of volume of endometriosis, there was no statistically significant difference. Conclusion Using the lancet to establish the endometriosis model has a high success rate, and subcutaneous model is better than peritoneum.

ABSTRACT

Considering the scarcity of studies on the behavior or reactivity of horses, this study aimed to assess the behavior of mares after insertion of intravaginal progesterone implant for use in cattle, through the observation of individual and collective behavior. In order to do so, seven mares received intravaginal devices impregnated with progesterone and luteolitic agent. After the implant was placed, the mares were released, and their behavior was observed (five observers with no contact between them) for a period of 120 minutes, following the model proposed by Heleski et al. (2002). The results indicated that during the 120-minute period, all mares were alert, in at least one observation; that all animals ate during the study period, and urination and defecation were observed in approximately 70 and 60%, respectively, of the assessed behavior. Behaviors that were likely an indication of pain and/or discomfort, like hitting the tail, miming to urinate, eversion of the clitoris and urinating were observed in up to 65 minutes, with the first three symptoms ceasing after 55 minutes. None of the analyzed behaviors were viewed after 65 minutes of observation. It can be concluded that the insertion of the intravaginal device impregnated with progesterone does not adversely affect the behavior of mares, and therefore, it can be used without compromising the well-being of the animals.
